Sai Tamhankar did her schooling from Sangli Convent School. She did her graduation in commerce from Sangli Chintaman College of Commerce. She was active in sports since her childhood and even though she was a Kabbadi player at a state level. Her enthusiasm towards acting rose when she was studied in college and during her college studies; she started participating in the stage functions and in the inter-college competitions.

 

She exhibited her acting talent in the play "Aadhe Adhure" and though she won the awards for Best Actress. She was more invigorated after getting this award. In 2008, she made her acting debut through the Subhash Ghai’s directed film Black & White starring Anil Kapoor, Aditi Sharma, and Shefali Chhaya. In the same following year, she appeared in the Marathi film Sanai Choughade as Sayee and in A. R. Murugadoss directed film Ghajini which was her career first ever-biggest hit and commercially gainer at the box office with the record collection of 232 crores.

 

In the subsequent time, she established herself as the lead heroines of the Marathi cinema. From 2008-18 she appeared in the max number of the Marathi films such as Mission Possible, Zhakaas, No Entry Pudhe Dhoka Aahey, Pune 52, Balak Palak, Time Please, Mangalashtak Once More, Guru Purnima, 3:56 Killari, Jaundya Na Balasaheb, and Rakshas. In those following year, she was cast in few Hindi films also City of Gold/Lalbaug Parel, Wake Up India and Hunterrr. Apart from acting she also hosted the shows such as Fu Bai Fu. And she acted in the serials such as  Kasturi, Agni Shikha, and Sathi Re.  She was highly appreciated for her performance in the film Love Sonia in which she was cast alongside Hollywood actress Demi Moore. She was recently seen in Marathi film Girlfriend which released in 2019.
