Soon, Lee also started writing comics and became the youngest comic editor at the age of 17. 1941 he published his first published work, a page of text in a Captain America comic, which he signed with the pseudonym Stan Lee. In the early 1950s, it was increasingly claimed that comics had a bad impact on young people; especially the popular horror comics were affected. To continue publishing, comic publishers carried out a self-censorship and as a result, created the strict comic code. In 1952, the DC comics Superman, Batman and Wonder Woman were the only regular titles.

In the late 1950s, DC Comics revived the superhero genre. The newly launched series The Flash and Justice League of America were very successful, and Martin Goodman commissioned Lee to create a new superhero team. Lee then created in 1961 with Jack Kirby the superhero family Fantastic Four, which were known in the German-speaking world as the Fantastic Four. After the successful launch of the series Lee and Kirby created shortly thereafter Hulk, Iron Man, Thor, and the X-Men. With Bill, Everett created Lee Daredevil and with Steve Ditko Doctor Strange and Spider-Man. These characters helped to reinvent the superhero genre.

In the 1960s, Lee was the author and editor of most Marvel series, responded to fan letters and wrote the monthly column "Stan's Soapbox”. In order to meet editorial deadlines, Lee invented his own style of comic. In 1981, he moved to California to promote Marvel's TV and film projects. In 2001, Lee first worked for DC Comics. He created the series "Just Imagine Stan Lee Creating”, a reinterpretation of various DC characters, as Lee would have created. For Spike TV Lee created the animated superhero series Stripperella. In 2004, plans were announced for collaboration with Hugh Hefner, which involved a superhero cartoon with Playboy playmates. In August 2004, Lee announced the formation of "Stan Lee's Sunday Comics," a website where subscribers can read Lee's new comics every Sunday.

In 2008, Lee received the National Medal of Arts. Since 2010, he has appeared as a presenter on the television series Stan Lee's Superhumans, which is broadcast in Germany under the title Superhuman on RTL II. In January 2011, Lee was on the Hollywood Walk of Fame with a star in the film category honoured.
