Sudhir Mishra was born and raised in Lucknow. He is the grandson of former Madhya Pradesh Chief Minister Dwarka Prasad Mishra. His father, Devendra Nath Mishra, was a founding member of the Lucknow Film Society. He graduated from Delhi University, where he met Badal Sircar and formed a theatre group called Workshop Theatre and worked on several plays with him. He then went to Pune and joined FTII. In 1980, he relocated to Mumbai and started working as an assistant director. He started off by writing script of the classic comedy film Jaane Bhi Do Yaaron in the year 1983. He then made his directorial debut through Bollywood superhit film Yeh Woh Manzil To Nahin in 1987. He ended up winning the Indira Gandhi Award for Best Debut Film of a Director. the following year, he won a National film award for his film Main Zinda Hoon. His film Dharavi also won a National Award. Off late, he has directed and worte screen play of films like Yeh Saali Zindagi, Inkaar, and Daas Dev.He has also served as the director of Hotstar web series Hostages in 2019.
His father's name was Devendra Nath Mishra. He has a brother named Sudhanshu Mishra. He was married to Sushmita Mukherjee and Renu Saluja in the past, He was also in a relationship with Bollywood actress Chitrangda Singh.
